At Beyond Insights, you’ll hear from Soumen Khan, PhD, Postdoctoral Researcher, Bioinformatics, Babraham Institute present his work determining the effect of Lin28a in differentiation using transcriptomics on the AVITI™. The temporal regulation of gene expression is instrumental to the proper development of an organism. However, despite its importance, the precise mechanism of biological timing is not fully understood. RNA binding protein (RBPs) are central in the dynamic control of the RNA life cycle and impact the overall rate of protein turnover through post-transcriptional regulatory mechanisms. Dr. Khan’s work aims to study how gene expressions change over time in the absence of an RNA-binding protein, Lin28a, and its implications in controlling the timing of development.

Abstract
Tephritidae is a highly speciated and globally distributed family of fruit flies that includes several species of economic interest due to their capacity to invade new environments and infest a wide variety of crops. Species identification is crucial for applying adequate measures of pest control and minimizing their economic impact. Most of the species catalogued as agricultural pests show similar morphology and have overlapping interspecific polymorphic characters. Furthermore, the popular COI marker has failed to accurately discriminate among most of these species. We have generated high quality genome assemblies that can be used as references for population genetics studies. Here, we present the first genome of Anastrepha fraterculus, which is a major pest in the Americas. We also applied whole genome sequencing technique to evaluate the mango fruit fly complex, which currently comprises four species (Bactrocera frauenfeldi, Bactrocera trilineola, Bactrocera caledoniensis and Bactrocera parafrauenfeldi) distributed in Southeast Asia and Oceania. B. frauenfeldi and B. trilineola are highly polyphagous and catalogued as major pests. We obtained a large set of SNPs, which were used to infer a robust phylogeny and evaluate their genetic differentiation. Based on this data, we have identified a set of markers that may be used for species discrimination, offering a foundation for developing more efficient diagnostic tools.
